The prion protein is not required for peripheral nerve de- and remyelination after crush injury

چکیده
The cellular prion protein (PrP) is essential to the long-term maintenance of myelin sheaths in peripheral nerves. PrP activates adhesion G-protein coupled receptor Adgrg6 on Schwann cells and initiates a pro-myelination cascade molecular signals. Because crucial for development regeneration after nerve injury, we investigated role repair. We performed experimental sciatic crush injuries co-isogenic wild-type PrP-deficient mice, examined repair processes. Generation cells, macrophage recruitment remyelination were similar mice. conclude that dispensable de- injury. may sustain its function independently activation by PrP.

Pioglitazone promotes peripheral nerve remyelination after crush injury through CD36 upregulation.
In our previous study, we found that CD36-deficient mice showed significant delays in peripheral nerve remyelination after sciatic nerve crush injury and suggested that CD36 played an important role in the restoration of injured peripheral nerves. The aim of this study was to investigate whether CD36 upregulation can promote peripheral nerve remyelination. We made crush injury that caused demye...
متن کاملAlphaB-crystallin regulates remyelination after peripheral nerve injury.
AlphaB-crystallin (αBC) is a small heat shock protein that is constitutively expressed by peripheral nervous system (PNS) axons and Schwann cells. To determine what role this crystallin plays after peripheral nerve damage, we found that loss of αBC impaired remyelination, which correlated with a reduced presence of myelinating Schwann cells and increased numbers of nonmyelinating Schwann cells....
متن کاملAxonally derived neuregulin-1 is required for remyelination and regeneration after nerve injury in adulthood.
Neuregulin-1 (NRG1) plays a crucial role in axoglial signaling during the development of the peripheral nervous system, but its importance in adulthood after peripheral nerve injury remains unclear. We used single-neuron labeling with inducible Cre-mediated knock-out animals, which enabled visualization of a subset of adult myelinated sensory and motoneurons neurons in which Nrg1 was inducibly ...
متن کاملPrion protein glycosylation is not required for strain-specific neurotropism.
In this study, we tested the hypothesis that the glycosylation of the pathogenic isoform of the prion protein (PrP(Sc)) might encode the selective neurotropism of prion strains. We prepared unglycosylated cellular prion protein (PrP(C)) substrate molecules from normal mouse brain by treatment with PNGase F and used reconstituted serial protein cyclic misfolding amplification reactions to produc...
متن کاملSpinal Microgliosis Due to Resident Microglial Proliferation Is Required for Pain Hypersensitivity after Peripheral Nerve Injury.
Peripheral nerve injury causes neuropathic pain accompanied by remarkable microgliosis in the spinal cord dorsal horn. However, it is still debated whether infiltrated monocytes contribute to injury-induced expansion of the microglial population. Here, we found that spinal microgliosis predominantly results from local proliferation of resident microglia but not from infiltrating monocytes after...
